  • Grapevine (Vitis vinifera) is one the most ancient and important fruit crop in the world. In late 2007 a major achievement has been reached in V. vinifera biology since the sequence of the grapevine genome has been published and it is available for public research. Thus, these facts prompted us to use the grapevine as a natural system to investigate the diversity of small RNAs expressed in different grape tissues (leaves, tendril, flower, berry) employing high-throughput sequencing (Solexa platform). In this work we have identified new grape specific miRNAs, which have tissue specific expression. The obtained sequencing data were analysed with bioinformatics approach and validated by northern blot analysis and in situ hybridization. Our analysis also confirmed the expression of several conserved miRNAs and reveals up to 20 novel grapevine-specific miRNAs. (literal)
