  • Phenol is a pollutant of surface and underground water, as it is present in industrial waste water. Under strict anaerobic conditions the denitrifying bacteria Thauera aromatica metabolise phenol to CO2 and H2O. The first step of the metabolic pathway is the phosphorylation of phenol to phenylphosphate that is then carboxylated to 4-hydroxybenzoic acid. The carboxylation involves two enzymes: phenol-kynase and phenylphosphate carboxylase. We have demonstrated that in vitro the phenylphosphate carboxylase enzyme catalyses the synthesis of 4-hydroxybenzoic acid starting from phenylphosphate and CO2 in mild conditions . 4-hydroxybenzoic acid, a species used for the production of innovative materials, is normally synthesised by the Kolbe-Schmitt reaction. We have demonstrated that the enzyme phenylphosphate carboxylase can be conserved in a active form in water solution for one or more days. We show that the enzyme can be used both under a high pressure in water solution or in SC-CO2. without any water. This is the first example of utilisation of a phenylphosphate carboxylase enzyme in SC-CO2. The selectivity of this method suggests the possibility of a large scale application, after optimization of the reaction system. A key step of the process is represented by the removal of the carboxylation product for avoiding its back conversion by the enzyme. In this perspective, we are working supporting the enzyme.
