- Ourmia melon virus (OuMV), the type member of the genus Ourmiavirus, has a tri-segmented plus-strand RNA genome. RNA1 and RNA2 encode a putative RNA-dependent RNA polymerase and a putative movement protein respectively. RNA3 encodes the 22k-Da coat protein (CP) which has no evident phylogenetic relationship to any protein in the database. Also the virion morphology determined by the CP is unique compared to other plant viruses. To better characterize the role of CP in OuMV life cycle, we have investigated the intracellular localization of green fluorescent protein-CP fusion (GFP-CP) via agroinfiltration into Nicotiana benthamiana leaves. Confocal laser scanning microscopy (CLSM) showed that GFP-CP was localized inside the nucleus and within the nucleolus in particular. We have identified the basic amino acid sequence 6KRKNRRNEKKK16 at the N-terminal of CP as a putative nuclear localization signal (NLS) and we are studying its role in CP targeting by progressive deletions and alanine scanning mutagenesis. The preliminary results showed that the first 16 amino acids are essential for the nucleolar localization of CP and the alanine substitution of Lys6 and Arg7 are sufficient to partially avoid nucleolar targeting of CP.
