- Ourmiaviruses belong to a genus of plant viruses which has only recently been characterized through the complete genome sequencing of its type member, Ourmia melon virus (OuMV). Ourmiaviruses have a trisegmented positive strand RNA genome coding for the RNA-dependent RNA polymerase (RdRp), the movement protein and the coat protein. None of them suppresses silencing, alone or in combinations, in a standard bioassay on 16C N. benthamiana transgenic lines for GFP, but the replicating wild type virus will suppress silencing around the inoculation foci, and during systemic infection. As a consequence of the infectious clone assembly, a mutant unable to cause the characteristic necrotic response in N. benthamiana was serendipitously originated. The same mutant is also severely impaired in silencing suppressor activity. We first mapped the determinant for the mild phenotype to the binary vector expressing the RNA1. Then a full length sequence comparison with a wild-type clone for RNA1 revealed a 1 nt deletion in a stretch of six cytosine residues in the coding region of the RdRP and 16 nt mutations distributed along the same RNA corresponding to three amino acids changes. Surprisingly, given the frame shift due to the c residue deletion, a symptomatic infection is established and the virus resulted from the infection restores the nt deletion to the wild type stretch of 6 cytosine residues. We are currently evaluating through viral chimeras and site directed mutagenesis which mutation is responsible for mild symptom expression and weak or absent silencing suppressor activity in the mild clone.
