  • Abstract: Aim of this work is the selection of rhamnolipid-producing strains that are able to synthesize monorhamnolipids or dirhamnolipids in preferential way. A panel of 39 isolates of P. aeruginosa was tested by a metabolic screening (methylene blue/CTAB agar plate method) and two strains, called L05 and L44, were selected as good rhamnolipids producers. To investigate the role of cultural conditions and carbon source, L05 and L44 were grown in two mineral media (Basal medium and E* medium, different for availability of N, Fe, Mg) and with two carbon sources: glycerol (three carbon atoms molecule) and Brassica carinata oil (very long chain fatty acids mixture). After 6 days fermentation, rhamnolipids were twice extracted with 2-fold volume of ethyl ether, were quantified by orcinol assay and were analyzed through 1 and 2D NMR spectroscopy. The data showed that in both tested media, only in reliance on carbon source, L05 exhibits a metabolic switch that allows to preferential production of about 80% of monorhamnolipids with Brassica carinata oil and 70% of dirhamnolipids with glycerol. L44, instead, produces a mixt are of mono- and dirhamnolipids, in all tested conditions. This work shows the possibility to produce mono-rhamnolipids through a fermentation process by a selected strain; in this way, is possible avoid the production of complex mixtures, decreasing strongly the costs need for rhamnolipids separation. Moreover, by only changing the carbon source, it is possible to direct the production of rhamnolipids form. (literal)
