  • The stem nematode, D. dipsaci ‘giant race’, is reported attacking broad bean in the Bekaa valley of Lebanon. The nematode, which is a major parasite of this crop and is widespread in many Mediterranean areas, colonised and reproduced within stems of field beans, invading the epidermis and several cell-layers of the cortical parenchyma. Brownish islands and distinct surface necrosis were visible mainly along the stem of infected plants, whereas cavities and necrotic areas, due to the trophic activity of the parasite, occurred in the cortex. All nematode developmental stages, including eggs, were observed in symptomatic tissues. Morphological and morphometrical data of adult specimens of D. dipsaci emerged from infected plant tissues are presented. PCR-RFLP analysis of the ITS containing region of ten individual nematodes was also carried out. The ITS product from three specimens was cloned and sequenced. Morphological and molecular approaches revealed our Lebanese population to be conspecific with Mediterranean giant races of D. dipsaci reported in literature on Vicia faba. (literal)
