Abstract
  • A protein inhibitor from Sulfolobulus Solfataricus and peptides thereof, which both have the ability to inhibit AARE and elastase are described. Similarly to the eukaryotic counterparts, the recombinant protein is able to inhibit in vitro the bovine alf a-chymotrypsin with a high specificity, and the porcine elastase but not all commercial available trypsins, features which distinguish all the members belonging to the family PEBP. Through site-specific mutagenesis techniques of the gene codifying SsCEI, it has been recognized the \"reactive site loop\" - RCL - on the inhibitor, responsible for the interaction with the eukarial protease target already identified. The inhibitor and the peptides thereof can be used as new compounds capable of modulating cognitive enhancement cardiovascular diseases, cancer, inflammation, hematological diseases, neurological diseases and urological diseases. (FR)La présente invention concerne un inhibiteur protéique provenant de Sulfolobulus Solfataricus et les peptides associés, possédant tous la capacité d'inhiber l'AARE et l'élastase. De même que ses homologues eucaryotes, la protéine recombinante est capable d'inhiber in vitro l'alpha-chymotrypsine bovine avec une haute spécificité, ainsi que l'élastase porcine, mais pas toutes les trypsines commercialement disponibles, caractéristiques qui distinguent tous les membres appartenant à la famille PEBP. Au moyen de techniques de mutagenèse dirigée du gène codant pour SsCEI, on a identifié la « boucle de site réactif » -RSL- sur l'inhibiteur, responsable de l'interaction avec la cible protéase eucaryote déjà identifiée. L'inhibiteur et les peptides associés peuvent être utilisés en tant que nouveaux composés capables de modulation dans le cas d'amélioration neurocognitive, de maladie cardio-vasculaire, de cancer, d'inflammation et de maladies hématologiques, neurologiques et urologiques.
