  • Arbuscular mycorrhizal (AM) symbiosis is an ancient and widespread mutualistic interaction involving most land plants (from liverworts to angiosperms) and a small group of obligate biotrophic soil fungi. The symbiosis improves plant mineral nutrition and fitness, thus having a crucial ecological importance. This chapter focuses on the events that allow AM fungi to move from the rhizosphere towards the root surface and eventually colonize the root cortex, where the main symbiotic functions develop. The process involves a series of events such as signaling and partner recognition, intraradical and intracellular fungal development and, last, the onset of nutrient exchanges that characterizes the functional symbiosis. Each step of this developmental programme is being investigated through a combination of approaches, ranging from genetics to transcriptomics, cell biology, proteomics and biochemistry. An overview on the most recent and significant achievements in these fields is presented in an attempt to outline a global view on the mechanisms that rule this ecologically essential – and scientifically fascinating – symbiosis.
