- Available data on the incidence and biodiversity of pome fruit viroids in Mediterranean basin are limited. Before starting a research survey to fill this gap, a tissue-printing hydridization (TPH) method to detect Apple scar skin viroid (ASSVd), Pear blister canker viroid (PBCVd) and Apple dimple fruit viroid (ADFVd) has been developed, validated and used in large-scale indexing of pome fruit viroids in Bosnia and Herzegovina, Malta, Lebanon and Turkey. A total of about 1,200 trees has been tested. Positive results obtained by TPH were confirmed by at least one additional detection method (RT-PCR and/or Northern-blot hybridization). PBCVd was detected in 18%, 12% and 8% of the tested pear trees in Bosnia and Herzegovina, Malta and Turkey, respectively, showing a wider diffusion of this viroid than expected. Interestingly, in all these countries several ancient native cultivars tested positive to PBCVd infection. In contrast, ASSVd was never detected and ADFVd was only detected in symptomatic trees (cv. Starking Delicious) in Lebanon, confirming a restricted spread of these viroids in Mediterranean basin. Full-length cDNA clones of PBCVd and ADFVd of the different geographic origin were molecularly characterized, with several new polymorphic positions in the genome of both viroids being identified. In the frame of this research, PBCVd in Bosnia and Herzegovina, Malta and Turkey, and ADFVd in Lebanon were first recorded, indicating that TPH is a useful technique for exploring pome fruit viroid spread.
