Satellite RNA (SatRNA) of Cymbidium Ringspot Virus (CymRSV) is a sub-viral RNA replicon found associated with its helper virus. It consists of a 621nt long RNA species sharing only a 50 nt long high similarity level sequence (termed Helper-Satellite Homology region-HSH) with the natural helper virus. RNA analysis of CymRSV-satRNA infected tissues revealed that the accumulation of satRNA considerably increases at low temperatures and/or in the presence of p19 silencing suppressor protein of helper virus. Moreover, the accumulation of two major satRNA truncated forms were observed, which likely are produced by the satRNA cleavage in the HSH region (position 230-231 nt). Another Tombusvirus member Carnation Italian ringspot virus (CIRV) can support the satRNA accumulation even at high temperature and in the absence of p19. The HSH region of CIRV displays lower similarity than that of CymRSV. By mutational analysis of CIRV we have shown that two mismatches occurring in the HSH sequence are responsible for the control of satRNA accumulation level. Last, we were able to detect the accumulation of the viral smalll RNA, which can programm RISC to satRNA targeting in the HSH region. The accumulation of satRNA targeting small RNA in CymRSV infected plants was also revealed by 454 high troughput paralel sequencing. These findings led us to conclude that CymRSV can activate RNA silencing to regulate the satRNA accumulation. (literal)
