Abstract
  • It has been estimated that more than 30 percent of human cancers can be prevented by appropriate diets. trans-resveratrol, a phytoalexin found in grapes and other plant products, was found to inhibit events associated with tumour initiation, promotion and progression, even though the fine-tuned molecular mechanisms mediating these effects are still unclear. MCF-7 cells were treated with various concentrations (ranging from 50 to 200 ?mol) of resveratrol for different time points (up to 48h). The effects of resveratrol on the cytoskeleton (re)organisation of MCF-7 were investigated by confocal microscopy using different fluorescent probes and semi-quantitative RT-PCR studying the expression of a set of genes specifically involved in such a physiological process. Finally, we used a 2D-GE (two-dimensional gel electrophoresis) approach to investigate main changes in the whole proteome of resveratrol-treated cells. Our results indicated that resveratrol inhibited tumour cell growth by inducing several cytoskeleton changes and finally apoptosis in MCF-7 cells. (literal)
