- Eleven populations of root-knot nematodes (RKN) have been collected from Venezuela and identified to species by means of isozyme electrophoresis patterns, obtained through mini-gels inserted into the automated equipment PhastSystemR. All the populations were previously reared on susceptible tomato in glasshouse. Adult females were extracted from the roots by cell-wall degrading enzyme mixture and sieving; collected under a stereoscope, and stored dry at -20°C. Samples for electrophoresis contained about 8 microg of nematode proteins. Gels were stained for 3 enzymes: esterase (EST), malate dehydrogenase (MDH) and superoxide dismutase (SOD). Three EST (II, El, M2), 3 MDH (N1, N1a, N2) and 2 SOD (N4, N4a) phenotypes were found indicating low variability among the populations collected. Most of the populations were from Aragua State and together with the population from Falcon State were identified as M. incognita. Only one population collected from coffee in Maracay (Aragua State) was identified as M. exigua. Isozyme patterns of two populations coming from Zulia and one from Lara State were ascribed to M mayaguensis. Search for possible minor mixed populations and tomato resistance-breaking isolates is still in progress. (literal)
