  • Purpose: Apoptosis plays a key role in the pathogenesis of heart failure (HF) contributing importantly to cell loss in the heart. Aim of this study was to assess the expression of apoptosis biochemical mediators in cardiac tissue of minipigs undergone pacing-induced HF, focusing attention on regional cardiac expression of apoptotic biomarkers, in order to better describe the apoptosis role in HF pathophysiology. Methods: Thirteen adult male minipigs (35--40 kg) were chronically instrumented with an unipolar pacemaker connected to the epicardial surface of the left ventricle (LV) anterior wall to induce HF by rapid LV pacing (180 beats/min). End-stage HF occurred at 21±2 days of pacing (LV end-diastolic pressure e20 mmHg). As control, 5 healthy adult male minipigs were also studied. mRNA expression of Caspase (Casp)-3, Casp-9 and their regulatory systems, Bcl-2 and Hsp72, respectively, were determined by semi-quantitative RT-PCR analysis in cardiac tissue from pacing site (PS) and opposite site (OS) wall of the LV. The apoptotic index was evaluated by TUNEL test. As inflammatory indices, Tumor Necrosis Factor (TNF)-a and Interleuchin (IL)-6 were also determined.
