  • Flood events may have long-term negative impacts on plant-soil systems critical for Citrus plants, causing premature senescence, which results in leaf chlorosis, necrosis, defoliation, cessation of growth and reduced yield. Citrus can be classified as a flooding-sensitive crop, although tolerance to this stress varies among genotypes and rootstocks. Carrizo citrange (Citrus sinensis x Poncirus trifoliata) is one of the most-used citrus rootstocks because of its high tolerance to the Citrus Tristeza virus, but it is quite sensitive to abiotic stresses, including oxygen deprivation. Based on our previous results demonstrating that the ectopic expression of the rice Mybleu transcription factor increases tolerance to oxygen deficit in Arabidopsis thaliana plants, we overexpressed Mybleu in Carrizo citrange to improve its tolerance to this stress. Through the evaluation of physiological, biochemical and molecular parameters, we found that Mybleu ectopic expression in Carrizo citrange increased tolerance to oxygen deficit, confirming the results obtained in Arabidopsis. At least in part this improved tolerance may be driven by changes in some biochemical and molecular parameters, such as the enzymatic activities of alcohol dehydrogenase and pyruvate decarboxylase as well as the expression levels of some genes responsive to hypoxic/anoxic conditions. Altogether our data suggest a key role of Mybleu in coordinating the multifaceted plant response to low oxygen stress and a conservation of the Mybleu-regulated pathways among species. (literal)
