  • The immunity system is the main barrier of the caprine mammary gland against bacterial infection and functions through the complex actions and interactions of hundreds of genes. Microarray analysis is a method developed to simultaneously study the expression of multiple genes in tissues as a response to a given treatment or physiological condition. The transcriptional mRNA expression of genes in milk somatic cells derived from goats with S. aureus mastitis were studied by means of microarray analysis. Gene expression analysis was performed using bovine cDNA microarrays containing about 10.000 genes. About 400 genes exhibited different expression during Staphylococcus aureus infection at the different time points post-infection. At the early stage of mammary infection in goats, Staphylococcus aureus induced a direct (lymphotoxin alpha, interleukin 1, alpha) and indirect (Xpo1 protein, CD2 antigen BP-2) release of cytokines, activation/inhibition of genes involved in control of inflammatory response and cytotoxicity (nitric oxide synthase 2A) and regulation of cell apoptosis (insulin-like growth factor BP 5, FOS-like antigen 2). The microarray data were then validated by real-time PCR. Further analysis of expressed genes involved in mammary infection with Staphylococcus aureus will improve the understanding of host response to mastitis and eventually aid in genetic selection of disease resistance.
