- Abstract
- I carcinomi tiroidei sono suddivisi in ben differenziati (papilliferi e follicolari), poco differenziati, anaplastici e midollari. Le alterazioni genetiche piu' frequenti sono:
1) attivazione oncogenica della chinasi RET mediante riarrangiamenti e mutazioni puntiformi nei tumori papilliferi e midollari, rispettivamente;
2) attivazione oncogenica della chinasi BRAF mediante mutazioni puntiformi nei carcinomi papilliferi ed anaplastici;
3) inattivazione di p53 nei tumori anaplastici.
Studieremo i pathways a valle di RET identificando nuovi substrati e il programma trascrizionale attivato da RET; valuteremo le variazioni nell'espressione genica tra carcinomi ben differenziati e anaplastici identificando i geni che partecipano alla \"proliferation and chromosomal instability signature\" tipica dei carcinomi anaplastici; analizzeremo la funzione biologica delle proteine CCDC6 e NCOA4 i cui geni sono riarrangiati con RET; studieremo come la proteina NFkappaB regola l'oncogenesi tiroidea; svilupperemo e valideremo nuovi tools in grado di bloccare l'attivita' di RET e di altre proteine oncogeniche dei tumori tiroidei.
