- Abstract
- Alcune proteine coinvolte nell'architettura cromatinica regolano a livello epigenetico la trascrizione di geni importanti per la trasformazione neoplastica. Tra queste le proteine HMGA, PATZ e CBX7. Studi recenti condotti dal nostro gruppo hanno dimostrato che il gene HMGA2, della famiglia dei geni HMGA, risulta essere amplificato e iperespresso in numerosi adenomi ipofisari e modelli animali iperesprimenti HMGA2 o HMGA1 sviluppano adenomi ipofisari secernenti prolattina e ormone della crescita. Tali risultati hanno suggerito un ruolo di queste proteine nella patologia oncologica ipofisaria. Il presente modulo di ricerca si propone di chiarire i meccanismi mediante i quali le proteine HMGA sono coinvolte nello sviluppo di adenomi ipofisari e nella tumorigenesi in generale. Tali meccanismi, una volta validati nel modello murino saranno estesi alla patologia umana. Inoltre, saranno sperimentati nuovi farmaci in grado di contrastare lo sviluppo e la progressione di tumori dipendenti dalle HMGA. Parallelamente sarà studiato il potenziale ruolo oncosoppressore di PATZ e di CBX7 mediante l'utilizzo di topi knockout, e il ruolo nella cancerogenesi umana di altre proteine cromatiniche. (literal)
